Recently, lab-on-handheld systems, such as microfluidic devices and Lab-on-a-Chip with smartphone (SP) imaging, have mainly been used to determine vitamins, minerals, and certain medications. In this study, a simple method is presented for accurately and sensitively measuring Captopril (CAP) using a spectrophotometer (spectroscopic reflectometer: SR) setup and a smartphone. The interaction between CAP and acid alizarin black dye (C26H16N4Na2O9S2) was evaluated at 35°C, using SR set to 510 nm wavelength. An iPhone 11 was used with a PC running ImageJ and Fiji software (Version 1.8.0). The linearity range of CAP concentration and RSD% was between 5 and 40 mg L-1 and less than 5%, respectively, by the SR method. The job method (JM), mole ratio (MR), conditional stability constant (Kf), Gibbs free energy (ΔG), and validation of the proposed system were studied. The application of this system for real sample analysis was compared to standard procedures. The innovative design showed excellent linearity, with correlation coefficients (r) of 0.9686, 0.9173, and 0.8635 for three model smartphones, measured using the Colorimeter (Red, Green, and Blue). The value of ΔG was found to be -3624.2, indicating the reaction’s spontaneity and supporting the confirmation of this study within the context of green chemistry. A strong positive correlation was observed between the new system, SR, and SP, as well as results from the SCIO-NIR micro-spectrometer device (handheld).
